Sex Determination by Polymerase Chain Reaction and Karyotyping of Bovine Embryos at First Cleavage in vitro

چکیده
The purpose of this study was to compare the embryo sexing methods between PCR and karyotyping of in vitro produced (IVP) bovine embryos at the 2-cell stage. The embryos were divided into 3 groups according to the time of first cleavage completion. Zygotes cleaving before 24 hours postinsemination (hpi), during 24-36 hpi and after 36 hpi were classified as fast, intermediate and slow cleaving embryos, respectively. All 2-cell stage bovine embryos were separated into two blastomeres, one was karyotyped and the other was subjected to PCR amplification. The percentages of embryos which were sexed by PCR were significantly (p<0.005) higher than those analyzed by karyotyping (100% vs 56%). The proportions of male embryos sexed by PCR and karyotyping in the fast and intermediate cleaving groups were significantly (p<0.05) higher than female embryos, whereas the proportions of males were significantly (p<0.05) lower in the slow cleaving group. The overall ratios of males to females analyzed by both methods were significantly (p<0.05) different from the expected ratio of 1:1.

Influence of selected (pre-)maturational parameters on in vitro development and sex distribution of bovine embryos.
The objectives of this research were to study the influence of a reduced oxygen concentration during in vitro maturation (IVM) and examine the effect of follicular glucose concentration on bovine in vitro development and sex distribution. In the first experiment, abattoir-derived cumulus-oocyte complexes (COC) were matured under 5% O2 or 20% O2. Secondly, COC were isolated and the glucose (G) c...
متن کاملP-50: Evolution of Bovine Embryos Cleavage in Co-Cultuer Mesenchyme Stem Cells (MSCs) Deried from Adipose Tissues
Background Mesenchyme stem cells (MSCs) are one of the widely used sources in tissue engineering and efficacy of cultural environment. In this survey, rat stem cells are used as coculture. Owing to main obstacle of embryo culture in the lab medium, the survival of embryo in lab is low in comparison with that of normal conditions. The purpose of this research is the survey of fertilization and c...
متن کاملLower quality bovine embryos may be successfully used for sex determination
The aim of this study was to evaluate the effect of sex determination procedures in Day 7 to Day 8 bovine embryos of various quality. For the purposes of comparison we used high quality (HQ) as well as lower quality (LQ) embryos obtained from superovulated donors. The healthy embryonic cells isolated from HQ embryos and blastomeres protruding into the perivitelline space of LQ embryos, were ana...
متن کاملAn Overview of Sex Selection at Conception in Mammals
Although the mechanisms under which sex ratio is controlled in nature remain unknown, researchers have attempted to treat sperm in an effort to predetermine the sex of the offspring. The methods of sperm separation that have been repeatedly reported to isolate fractions rich in X- or Y-chromosome bearing sperm are serum albumin gradient swim-up and flow cytometry. However, the ability of the se...
متن کاملIn vitro and in vivo quality of bovine embryos in vitro produced with sex-sorted sperm.
In this work we analyzed the effects of three culture systems on developmental ability of bovine embryos in vitro produced with sexed sperm, the survival to vitrification (cryologic vitrification method) of such blastocysts, and their pregnancy rates after embryo transfer to recipients, both as fresh and after vitrification/warming. Finally, we measured the accuracy of the sorting protocol by a...
